Welcome to the Drupal community. Unfortunately, your question is too broad and would require too much hand holding. I wouldn't expect anyone here in these forums to take you through the entire process of building a site like 99 designs. Start by getting to know Drupal http://drupal.org/documentation , then open a new thread for your questions.

Also, you may want to edit your initial post as you have tagged it with "Deprecated - Database compatibility", which as it says it is deprecated and also not applicable to what you are trying to do.
