Dear Omega 4 Team,

i make a Subtheme and everything is fine.

But now a made a new regions, make a entry in the ../theme/layouts/my-subtheme/my-subtheme.inc and also by my_subtheme.tpl.php, clear the cache 4x and nothing.
What is wrong?

You have to add the new region to both, your theme's .info file and the yourlayout.layout.inc file. Otherwise it won't be available for your .tpl.php
