Confused

I'm pretty new to Drupal, so if this is a stupid question, sorry.
What exactly do I do here: set the DRUPAL_ROOT variable at the top of the adserve.inc file?

EX.: //define 'DRUPAL_ROOT', '/var/www/ "mysite?"' ;

Or, something else?
