The scenario is:

Drupal 5.7
jQuery Update 5.x-2.x-dev (2008-Jun-08) - Also tested with RC1.
jQuery plugins 5.x-1.3
jQuery Map Hilight 5.x-1.x-dev (2008-Abr-24)
Javascript Tools 5.x-1.1 (I mention it, just in case is there any lib conflict... for instance with collapsiblock.js).

README instructions read and strictly followed (misc drupal files replaced with jquery_update and jquery.maphilight.min.js copied to jquery_plugins).

Modules were enabled (one by one) with any reported error or warning.

When I create a new page with the test image (as is said in readme) I got a:

N.metadata is not a function
http://psicologiasocial.uab.es/colaborando/sites/all/modules/jquery_plugin/jquery.maphilight.min.js
Line 1

Any clue about the culpit of this issue?

Are you still having this problem? I was unable to replicate it on a fresh install as follows:
