I know the main idea of layers with Drupal, but looking throught the massive list of building links I am lost.

how to set up a local test area on your local machine depends on what software you choose to use.
There are many LAMP stacks available for both Mac and Windows.
http://drupal.org/contributors-guide is a good place to start armed with google to research the different setups available.

This thing is awesome
It even allows to set-up multisites which is awesome to quickly test different settings. I highly recommend it for testing and breaking. ;)
