I'm a Drupalnewbie and couldn't get Lightbox2 working with a fresh installed D7. I did the following:
I added an Image field to Article and Basic content type as described here: https://drupal.org/documentation/modules/image
Lightbox Lite is DISABLED (by default).
In section admin/config/user-interface/lightbox2/automatic for "Automatic handler for image nodes" I selected Lightbox.
According to my understanding this is all which I need to configure to get Lightbox2 working. However, when viewing Articles or Basic contents with uploaded images, no Lightbox2 appears. Any advice is greatly appreciated. Please let me know.

stefanx CreditAttribution: stefanx commentedI discovered that in section /admin/structure/types I had to select Manage Display for each content type and for the Image field select a Lightbox2 entry in the Format pull-down menu. Now it works. :-)

I'm using the flickr module, and I was trying to get it so clicking on an image in the photoset would open in a lightbox. It is adding the tags needed to the images, but it's just not working.

lolandese CreditAttribution: lolandese commentedThis is since a while natively supported from the side of the Flickr module. See #1637464: Flickr: Colorbox/Lightbox integration.. It works for any overlay browser (also Colorbox) that gets triggered by the 'class' and/or 'rel' attribute of the anchor link that wraps around an image.
