How to sort profile settings, disable some fieldsets, etc.

Not at the moment. You can hide them I think in Content Profile settings.

Another issue was asking for weight configuration to show the Profile fields above the Account fields: #1121534: Assigning Weights.

I posted there too that you could probably do a custom module and use hook_form_alter() to change the fields' weights. Although, it would be much nicer if this module had a UI feature to do this!
