Lightbox2 has already been updated to work with imagecache V2 (still not officially released btw). However, while testing this, I found a separate issue with viewing imagecache images in the lightbox and so have applied a couple of patches to CVS for this. These will be included in the next release and will also be in the next dev release (available later today).
Both lightbox2 5.x and 6.x versions support both versions of imagecache. There's no 6.x version of imagecache yet, but lightbox2 6.x assumes that the api hasn't changed since the 5.x version. Once a imagecache 6.x version is available, I'll re-test this.
If you use the latest dev version you can switch to using imagecache v2 and new lightbox - I did find an issue with the imagecache support as mentioned in comment #1. Though, I would recommend testing on a non-production site first as always :)
I can vouch for the lightbox2 dev code being stable - it's certainly no worse than the last official release! However, I can't say one way or another about the imagecache2 code, not that I doubt the ability of its maintainers - it's more of a "if it's still beta, then it's beta for a reason" sorta thing.
