The weight markup settings are disabled and cannot be changed. These settings are in Store > Configuration > Shipping Quotes > USPS/UPS Configure > Markups

I'm running the latest version of Drupal core and everything else seems to be working fine. Anyone else have this problem?

That option is deliberately disabled for UPS and USPS because it is not yet fully implemented. (It IS implemented for uc_flatrate and uc_weightquote.) You can help out with #943778: UPS Weight Markup if this is important to you.
