In a productive multi-site-system with seven sites i am using successful plupload even in combination with Media Browser Plus (MBP). Thanks to all the contributors.
But now, suddenly, in ONE of seven sites the Plupload-Widget is not displayed anymore.
I don't know why because I didn't change someting using the media stuff.
[Supplement:] And even an older version of the site with less media files and nodes on my local Mac has the same problem, but not the other sites using the same stuff (libraries, modules, even themes etc.).
I did control the permissions (i am number one).
I compared the enabled modules (with Drush and Excel Lookup)
I controlled all media & file settings
I controlled the theme settings (same admin theme)
I toggled enabling/disabling plupload (the Single file upload form element does appear)
I rebuild permissions und started cron jobs.
[Supplement:] I also installed the plupload.module.patch for Drupal 7.23 (even 7.28 is the current version). Same Issue, other sites not affected.
[Supplement:] Just one of seven sites (my private site :-( ) has the problem with the broken plupload integration on Debian and on my Mac.
[Supplement:] I also changed the theme.
With Plupload enabled the form containers (div) for plupload is outputted but empty.
For sure, everything in this multisite system is up to date.
Do you have any hint? Your answer will be fully appreciated.

I solved the problem on my own, but thanks for your attention.
Even I am more conservative in technical aspects the default version of jQuery was set to 1.10 and not on 1.8 in the «jQuery Update»-Settings, which is apparently correct.
Firebug in Safari delivered the indication.
