Is there a way to change the layout/template for a page once I've published it? Or even better, a way to add blocks to a page post publishing?
Sorry if this is something basic, but I've been unable to find an answer anywhere.

Blocks can be set in administer -> blocks. Blocks are indenepent of nodes and can be set pre/post publishing of cotent.
As far as layout/template, please clarify? in general the answer is yes.

If I've created a "featured story" template in addition to my "basic" template, can I apply my "featured story" template to a node that I had previously published with a "basic" template?

I assume that when you state "template" you mean tpl.php file. note: the theme layer and content are completely separate. You can change the template for any part of the site at any time.
